So far, 6 types and more than 20 subtypes of crispr-Cas systems have been identified. Among them, CRISPR-Cas12a is a class II (V-type) CRISPR-Cas system for editing mammalian genomes. Cas12a has unique features that can be complementary to the CRISPR-Cas9 system. The Cas12a (Cpf1) protein has a RubC endonuclease domain similar to Cas9. In addition, Cpf1 does not have the HNH endonuclease domain, and the N-terminal of Cas12a does not have the α-helix recognition leaf of Cas9.
Research shows that the binding of lbCas12a-crRNA complex to complementary single-stranded OR double-stranded DNA releases strong non-specific ssDNA trans-cleavage activity. This has prompted Cas 12a to be used as a new type of tool for genetic testing and imaging.
Features:
Cpf1 does not require traceRNA, only crRNA, which is very beneficial for genome editing; Cas12a is smaller than Cas9 and also has a smaller crRNA molecule (nearly half of Cas9's total sgRNA); The Cas12a-crRNA complex cleaves target DNA by recognizing a T-rich PAM motif, in contrast to the G-rich PAM required by cas9.
