Product Description:

Recombinant TEV protease is obtained by genetic engineering expression and purification, and TEV protease specifically recognizes the seven amino acid sequence consisting of Glu-Asn-Leu-Tyr-Phe-Gln-Gly. Compared with EK, SUMO and other proteases, it has the characteristics of high activity and high specificity. TEV protease is purified by 6×His tag (including histidine tag), and its purity is as high as 99%; after the enzyme cleavage reaction is completed, It can be removed by the His-tag purification resin Ni-NTA Resin, so as to achieve the purpose of purifying the target protein. It has activity under the reaction conditions of 4-30℃ and pH 6.0-8.5, and the relative reaction conditions are relatively loose.

Instructions:

Before use, pre-enzyme digestion of 10μg-30μg protein sample can be used to determine the optimal reaction conditions and optimal enzyme dosage. For proteins that are not easy to be digested, the enzyme amount can be appropriately increased and the reaction time can be extended. Recommended digestion temperature: 4°C-25°C, overnight at 4°C or 1 hour at room temperature.

Precautions:

PMSF and AEBSF (1 mm), TLCK (1 mm), bestatin (1 mg/ml), pestatin a (1 mm), EDTA (1 mm) and E-64 (3 mg/ml). The above protease inhibitors had no effect on TEV activity; Zn ion (> 5 mm) inhibited TEV activity; Reagents that react with cysteine are also potential inhibitors of TEV.
