CRISPR-Cas9 is an adaptive immunedefense formed by bacteria and archaea in the long-term evolution process, whichcan resist the invading virus and foreign DNA. It can integrate the fragmentsof invading phage and plasmid DNA into CRISPR sequence, and guide thedegradation of homologous sequence of cas9 protein through corresponding CRISPRRNAs (crRNAs), so as to provide immunity. CRISPR-Cas9 can be used for in vitrocleavage, gene editing of living cells, and optical probe biological markers.Cas9 is stably combined with gRNA in vitro, and the Cas9/gRNA complex can be directly cut into the cell genome by transfectioninto the cell through electrotransfer, liposome transfection reagent, or a newnano vector. Cas9 NLS can also be transferred into cells together with gRNAexpression plasmids and recombinant templates to realize site-specific editingof cell genome.
